چکیده

The Wnt/β-catenin signaling pathway plays a key role during embryonic development, homeostasis and cancer. of β-catenin in melanocyte lineage establishment has been elucidated, but its melanoma remains poorly defined. In particular, function cell invasion is controversial. Especially, we generated signature activation to emerge proteins interest including known target genes (AXIN2, APCDD1, NKD1, SP5, NOTUM, ZNRF3, CCND1 PPARD) new (SLC1A5, SLC7A11, SLC24A4 MICAL2). Our gene most correlated with invasive cells MICAL2. MICAL2 oxidizes actin filaments, destabilizes them, modifies the migratory power cells. lines, reduction or induction reduces increases level MICAL2, respectively. Decreasing migration, growth. Finally, significantly unfavorable evolution for patient associated strong expression power. conclusion, identification transcriptional an important step towards understanding melanomagenesis. which pharmacologically targetable example.

Silymarin Targets β-Catenin Signaling in Blocking Migration/Invasion of Human Melanoma Cells

Metastatic melanoma is a leading cause of death from skin diseases, and is often associated with activation of Wnt/β-catenin signaling pathway. We have examined the inhibitory effect of silymarin, a plant flavanoid from Silybum marianum, on cell migration of metastasis-specific human melanoma cell lines (A375 and Hs294t) and assessed whether Wnt/β-catenin signaling is the target of silymarin. U...

Upregulated MicroRNA-25 Mediates the Migration of Melanoma Cells by Targeting DKK3 through the WNT/β-Catenin Pathway

Previous research indicates that microRNA-25 (miR-25) regulates carcinogenesis and the progression of various cancers, but the role of miR-25 in melanoma remains unclear. We observed that miR-25 was significantly upregulated in melanoma cell lines and tissue samples. Downregulation of miR-25 markedly suppressed invasion and proliferation of melanoma cells in vitro; however, overexpression of mi...

RTL1 promotes melanoma proliferation by regulating Wnt/β-catenin signalling

Cutaneous melanoma is a highly malignant and metastatic skin cancer with high mortality. However, its underlying mechanisms remain largely unclear. Here, we found that retrotransposon-like 1 (RTL1) is highly enriched in melanoma tissue, especially in early and horizontal growth tissues. Knockdown of RTL1 in melanoma cells resulted in cell proliferation suppression; cell cycle arrest at G1 phase...

β-Catenin promotes cell proliferation, migration, and invasion but induces apoptosis in renal cell carcinoma

β-Catenin (CTNNB1 gene coding protein) is a component of the Wnt signaling pathway that has been shown to play an important role in the formation of certain cancers. Abnormal accumulation of CTNNB1 contributes to most cancers. This research studied the involvement of β-catenin in renal cell carcinoma (RCC) cell proliferation, apoptosis, migration, and invasion. Proliferation, cell cycle, and ap...

WLS inhibits melanoma cell proliferation through the β-catenin signalling pathway and induces spontaneous metastasis

Elevated levels of nuclear β-catenin are associated with higher rates of survival in patients with melanoma, raising questions as to how ß-catenin is regulated in this context. In the present study, we investigated the formal possibility that the secretion of WNT ligands that stabilize ß-catenin may be regulated in melanoma and thus contributes to differences in ß-catenin levels. We find that W...
